Detecting genetic hypermutability of gastrointestinal tumor by using a forensic STR kit

摘要: Growing evidence suggests that somatic hypermutational status and programmed cell death-1 overexpression are potential predictive biomarkers indicating treatment benefits from immunotherapy using immune checkpoint inhibitors. However, biomarker-matched trials are still limited, and many of the genomic alterations remain difficult to target. To isolate the potential somatic hypermutational tumor from microsatellite instability low/microsatellite stability (MSI-L/MSS) cases, we employed two commercial kits to determine MSI and forensic short tandem repeat (STR) alternations in 250 gastrointestinal (GI) tumors. Three types of forensic STR alternations, namely, allelic loss, Aadd, and Anew, were identified. 62.4% (156/250) of the patients with GI exhibited STR alternation, including 100% (15/15) and 60% (141/235) of the microsatellite high instability and MSI-L/MSS cases, respectively. 30% (75/250) of the patients exhibited STR instability with more than 26.32% (26.32%–84.21%) STR alternation. The cutoff with 26.32% of the STR alternations covered all 15 MSI cases and suggested that it might be a potential threshold. Given the similar mechanism of the mutations of MSI and forensic STR, the widely used forensic identifier STR kit might provide potential usage for identifying hypermutational status in GI cancers.

摘要: A large number of expressed sequences tags are available for spp., which provides an opportunity to understand genomic organization of the transcribed regions. Here, we report a detailed analysis of repetitive elements including tandem repeats (TRs) and transposable elements (TEs) in the transcribed region of the spp. On average, 22% of the expressed sequence tags (ESTs) contain TRs. The relative density of TR classes is highly taxon-specific. For instance, has a high relative density of mononucleotide repeats, whereas dinucleotide repeats are rare. The proportions of 2–6, 7–30 and 31–50 bp repeats were almost identical in all studied species except for and . We found that<1% of the citrus ESTs have a similarity with transposable elements. Transcriptional activity of transposable element families varied even within the same class of elements. A high proportion of transcriptional activity was observed for -like TEs compare to other TE classes. While TEs are relatively rare, TRs are abundant elements in ESTs of citrus. The high proportion of TRs that have a unit size longer than 6 bp raises the question about a possible functional or evolutionary role of these elements.

Alluvial channel hydrodynamics around tandem piers with downward seepage

摘要: In this paper, we report the turbulent flow structures and the scour geometry around two piers with different diameters. An experiment was conducted on a non-uniform sand bed with two types of tandem arrangements, namely, pier (T1) with a 75 mm front and 90 mm rear, and pier (T2) with a 90 mm front and 75 mm rear, with and without-seepage flows, respectively. A strong wake region was observed behind the piers, but the vortex strength diminished with downward seepage. Streamwise velocity was found to be maximum near the bed downstream of the piers and at the edge of the scour hole upstream of the piers. Quadrant analysis was used to recognize the susceptible region for sediment entrainment and deposition. Upstream of the piers near the bed, the moments, turbulent kinetic energy (TKE), and TKE fluxes were found to decrease with downward seepage, in contrast to those in a plane mobile bed without piers. The reduction percentages of scour depth at the rear pier compared with the front one were approximately 40% for T1 and 60% for T2. Downward seepage also resulted in restrained growth of scouring with time.

Design of tandem genes cluster for isoflavone engineering

摘要: In this paper, a universal effective novel method of constructing tandem genes cluster for isoflavone engineering was reported. A tandem genes cluster () of secondary metabolites of plant isoflavones was constructed by using the chalcone synthase (CHS), chalcone isomerase (CHI), and isoflavone synthase (IFS) (GenBank accession numbers EU526827, EU526829, EU526830) in only one recombination with the pET22b vector. The resulting expression vector pET- was heterogeneously expressed and co-incubated with barrenwort extractions, and the genistein-like component was detected.

Available energy analysis of new tandem double-capillary tube refrigeration system for refrigerator-freezers

摘要: A new tandem double-capillary tube refrigeration system for refrigerator-freezers is proposed. A capillary tube was added between the two evaporators in the fresh and frozen food storage chests to raise the evaporation temperature of the refrigerating chamber, and reduce the heat exchange temperature difference and the available energy loss. Peng-Robinson (P-R) equation of state was adopted to calculate the thermodynamic properties of the refrigerants, and the available energy analysis of the vapor compression refrigeration cycle was programmed to calculate the thermodynamic performances of the new and the conventional refrigeration cycle of the refrigerator-freezer. The calculation results show that the available energy efficiency of the conventional refrigeration cycle of the refrigerator-freezer is 21.20% and 20.57%, respectively when the refrigerant is R12 and R134a, while that of the double-capillary tube refrigeration cycle of the refrigerator-freezer is 23.97% and 23.44%, respectively. By comparison, the available energy efficiency of the new refrigeration system increases by 13.07% and 13.95%, respectively.

quantification of several classes of antibiotics in water, sediments, and fish muscles by liquid chromatography–tandem

摘要: Precise and sensitive methods for the simultaneous determination of different classes of antibiotics, including sulphonamides, fluoroquinolones, macrolides, tetracyclines, and trimethoprim in surface water, sediments, and fish muscles were developed. In water samples, drugs were extracted with solid-phase extraction (SPE) by passing 1000 mL of water through hydrophilic lipophilic balanced (HLB) SPE cartridges. Sediment samples were solvent-extracted, followed by tandem SPE (strong anion exchange (SAX) + HLB) clean-ups. Fish muscles were extracted by a mixture of acetonitrile and citric buffer (80:20, v/v) solution, and cleaned by SPE. Liquid chromatography–tandem mass spectrometry (LC-MS/MS) with multiple reaction monitoring (MRM) detection was employed to quantify all compounds. The recoveries for the antibiotics in the spiked water, sediment, and fish samples were 60.2%–95.8%, 48.1%–105.3%, and 59.8%–103.4%, respectively. The methods were applied to samples taken from Dianchi Lake, China. It showed that concentrations of the detected antibiotics ranged from limits of quantification (LOQ) to 713.6 ng·L (ofloxacin) in surface water and from less than LOQ to 344.8 μg·kg (sulphamethoxazole) in sediments. The number of detected antibiotics and the overall antibiotic concentrations were higher in the urban area than the rural area, indicating the probable role of livestock and human activities as important sources of antibiotic contamination. In fish muscles, the concentration of norfloxacin was the highest (up to 38.5 μg·kg ), but tetracyclines and macrolides were relatively low. Results showed that the methods were rapid and sensitive, and capable of determining several classes of antibiotics from each of the water, sediment, and fish matrices in a single run.

摘要: A rapid, inexpensive and laboratory friendly method was developed for analysis of off-flavor/odor compounds in fresh and salt water using gas chromatography with chemical ionization-tandem mass spectrometry. Off-flavor/odor compounds included geosmin, 2-methylisoborneol (MIB), 2-isobutyl-3-methyoxypyrazine (IBMP), and 2-isopropyl-3-methoxypyrazine (IPMP). Using this method, a single sample can be extracted within minutes using only 1 mL of organic solvent. The ion transitions for IPMP, IBMP, MIB, and geosmin were 153>121, 167>125, 152>95, and 165>109, respectively. The linearity of this method for analyzing MIB ranged from 4 to 200 ng·L , and from 0.8 to 200 ng·L for the other analytes. Method recoveries ranged from 97% to 111% and percent relative standard deviations ranged from 3% to 9%, indicating that the method is accurate, precise, and reliable.

Study on the crystal morphology and melting behavior of isothermally crystallized composites of short

摘要: The spherulites of the short carbon fiber(SCF)/poly (trimethylene terephthalate) (PTT) composites formed in limited space at designed temperatures, and their melting behaviors were studied by the polarized optical microscopy, atomic force microscopy (AFM), and scanning electron microscopy (SEM), respectively. The results suggest that SCF content, isothermal crystallization temperatures, and the film thicknesses influence the crystal morphology of the composites. The dimension of the spherulites is decreased with increasing SCF content, but whether banded or nonbanded spherulites will form in the composites is not dependent on SCF content. However, the crystal morphology of the composites depends strongly on the temperature. When the isothermal crystallization temperatures increase from 180°C to 230°C, the crystal morphology of SCF/PTT composites continuously changes in the following order: nonbanded → banded → nonbanded spherulites. Discontinuous circle lines form in the film when the film thickness increases from 30 to 60 μm. Basing on the SEM observation, it is found that these circle lines are cracks formed due to the constriction difference of the different parts of the spherulites. These cracks are formed when the film is cooled from the isothermal crystallization temperature to the room temperature at a slow cooling rate; while they will disappear gradually at different temperatures in the heating process. The crack will appear/disappear first around the center of the spherulite when the film was cooled/heated. The nontwisted or slightly twisted lamellas will reorganize to form highly twisted lamellas inducing apparent banded texture of the spherulites.

摘要: Combined with the actual project, this paper carries out a nonlinear finite element analysis on 2 groups, 6 short-limbed shear walls, through the finite element calculation software ANSYS. The stress-strain relation of the models, and the effects of the type of sections and the axial compression ratios on the models can be obtained, providing a reference for future design.

Ultrathin microcrystalline hydrogenated Si/Ge alloyed tandem solar cells towards full solar spectrum

摘要: Thin film solar cells have been proved the next generation photovoltaic devices due to their low cost, less material consumption and easy mass production. Among them, micro-crystalline Si and Ge based thin film solar cells have advantages of high efficiency and ultrathin absorber layers. Yet individual junction devices are limited in photoelectric conversion efficiency because of the restricted solar spectrum range for its specific absorber. In this work, we designed and simulated a multi-junction solar cell with its four sub-cells selectively absorbing the full solar spectrum including the ultraviolet, green, red as well as near infrared range, respectively. By tuning the Ge content, the record efficiency of 24.80% has been realized with the typical quadruple junction structure of a-Si:H/a-Si Ge :H/µc-Si:H/µc-Si Ge :H. To further reduce the material cost, thickness dependent device performances have been conducted. It can be found that the design of total thickness of 4 m is the optimal device design in balancing the thickness and the . While the design of ultrathin quadruple junction device with total thickness of 2 m is the optimized device design regarding cost and long-term stability with a little bit more reduction in . These results indicated that our solar cells combine the advantages of low cost and high stability. Our work may provide a general guidance rule of utilizing the full solar spectrum for developing high efficiency and ultrathin multi-junction solar cells.

Experimental investigation and comparative study of inter-turn short-circuits and unbalanced voltage

摘要: A transient model for an induction machine with stator winding turn faults on a single phase is derived using reference frame transformation theory. The negative sequence component and the 3rd harmonic are often considered as accurate indicators. However, small unbalance in the supply voltage and/or in the machine structure that exists in any real system engenders the same harmonics components. In this case, it is too difficult to distinguish between the current harmonics due to the supply voltage and those originated by inter-turn short-circuit faults. For that, to have the correct diagnosis and to increase the sensitivity and the reliability of the diagnostic system, it is crucial to provide the relationship between the inter-turn short-circuits in the stator winding and the supply voltage imbalance through an accurate mathematical model and via a series of experimental essays.

Anlotinib as third- or further-line therapy for short-term relapsed small-cell lung cancer: subgroup

摘要: Patients with small-cell lung cancer (SCLC) relapse within months after completing previous therapies. This study aimed to investigate the efficacy and safety of anlotinib as third- or further-line therapy in patients with short-term relapsed SCLC from ALTER1202. Patients with short-term relapsed SCLC (disease progression within 3 months after completing ≥ two lines of chemotherapy) in the anlotinib (n = 67) and placebo (n = 34) groups were analyzed. The primary endpoint was progression-free survival (PFS). The secondary endpoints included overall survival, objective response rate (ORR), disease control rate, and safety. Anlotinib significantly improved median PFS/OS (4.0 vs. 0.7 months, P < 0.0001)/(7.3 vs. 4.4 months, P = 0.006) compared with placebo. The ORR was 4.5%/2.9% in the anlotinib/placebo group (P = 1.000). The DCR in the anlotinib group was higher than that in the placebo group (73.1% vs. 11.8%, P < 0.001). The most common adverse events (AEs) were hypertension (38.8%), loss of appetite (28.4%), and fatigue (22.4%) in the anlotinib group and gamma-glutamyl transpeptidase elevation (20.6%) in the placebo group. No grade 5 AEs occurred. For patients with short-term relapsed SCLC, third- or further-line anlotinib treatment was associated with improved survival benefit. Further studies are warranted in this regard.
